Adeno-associated virus (AAV) vectors have emerged as a potent tool to deliver genes in research and for clinical treatment via gene therapy. We describe a method to measure the number of incoming AAV genome co-expressed per cell. This paper provides a general protocol for A) construction and production of triple-AAV vectors, B) AAV infection in cell culture and AAV injection in the mouse, C) image analysis and AAV genome quantification with single-cell resolution.
